As far as scales and the like go maybe Kenny will chime in with his perspective.
I play predominantly blues pentatonic with major mixolydian accents. Note on my post where I cite the chorus lick. This is a major scale phrase. You could play major throughout but it won't have the edge you're probably looking for.
Most of the nasty stuff is JG leaning on the blue notes with a small dose of chromaticism. Jerry often moved seamlessly between major and blues/minor phrasing which gives great contrast.
